Sélection de zones racinaires de plantules de luzerne pour les préparer afin de les observer en microscopie à champ large, en lumière blanche ou fluorescence. Il s’agit d’une culture in vitro de plantules de luzerne, "Medicago truncatula", en milieu gélosé, infectées par un champignon, "Aphanomyces euteiches", avec dépôts de spores au niveau de la racine. L’objectif est de localiser à l'échelle cellulaire le champignon dans la racine de luzerne (intra ou extracellulaire).
